Use RowDataBind event to get access to the cell and add content to the tooltip

you can do it in one more easiest way .. using label... like this..

<asp:templatefield>
<itemtemplate>
<asp:label runat="server"
text='<%# Eval ( "ProductID" ) %>'
tooltip='<%# Eval ( "ProductDescription" ) %>' />
</itemtemplate>
</asp:templatefield>
